Cryopreservation of Hematopoietic Stem-Cell Product utilizing DMSO- Preparation of CAR-T Products- Flow Cytometry Analysis (BD)o Flow Cytometry was utilized to determine Stem-cell dose of product using CD-34 anti-body. o When performing a fresh infusion from a related- or un-related donor, flow analysis is performed to determine stem-cell & T-cell dosage. The latter was performed to ensure T-Cells dosage is within Standard Operating Procedure utilizing CD-3 Anti-body- Hemocytometry analysis (Sysmex)o Utilized as a 2nd verifier to validate the results from flow analysis- Preparation of Fresh Stem-Cell Product from Donor following established Standard Operating Procedure: o Donor Status: Un-related, Related, & Haplo-o Product Partitioning Product partitioned based on TNC of acquired product, and/or per physician request. o Cryopreservation of Product not infused after fresh infusion. Prepared in the form of DLI’s, partitioned by requested dose from the physician- Experience in the utilization of Microsoft Excel and Power Bi for functions related to the lab and record keeping. Microsoft Excel was used to produce automated lab sheets that functioned to perform electronic calculations that were otherwise done manually. This action increased workflow efficiency, also decreasing calculation error. Every sheet produced for different forms of processing was put through an extensive validation process, later approved by the leadership amongst the cryo-lab. Microsoft Excel was also utilized as a data-base platform. It functioned to produce dynamic visuals that would illustrate how many products could be produced in a year. As well as providing visuals for other forms of data related to time of engraftment post infusion, and how many infusions could occur in a quarter-to-a-year period. Microsoft Power Bi is a presentation-platform that can be used to provide more in-depth visuals from alternate data-base platforms. This presentation-platform has proven to provide more dynamic visuals than have been provided by Excel.
