Background

About Anup Parchure

I am currently pursuing my Post-doctoral research at Yale school of Medicine. I have led a project on deciphering the role of LLPS in protein sorting from the Golgi apparatus for physiologically relevant molecules like insulin. I have built up on my previously acquired skills and developed new skills like protein purification by establishing for the first time purification of tagged versions of Secretory Granule forming proteins in their native form and used it for phase separation assays. Standardized in vivo experiments to demonstrate phase separation in the Golgi lumen for biogenesis of secretory granules and delivery of physiologically relevant molecules like Proinsulin. This fundamental discovery connecting principles of protein secretion to Diabetes Research has been published and highlighted in Science journal and also found it's way in a book chapter. I have also co-authored another manuscript which is currently under review and another manuscript under preparation Established extensive collaborations with research groups across US and UK. Funded by the Yale Diabetes Research core, I am establishing the role of condensates in biology of Adipocytes for regulation of insulin signaling. Previously I did a short post doc at UMASS medical school in the Budnik laboratory in the department of Neurobiology and I characterized a role for non canonical RNA export via nuclear envelope budding in mammalian cell types, which is necessary for maintenance of mitochondria and development of neurons. During my PhD with Jitu Mayor, at the National Center for Biological Sciences, I worked at the interface of cell and developmental biology and uncovered the mechanisms underlying transport of Hedgehog across tissues. We uncovered an exosomal means of Hedgehog transport in the tissue which was critical for it's long range signaling. This was published in Molecular Biology of the Cell. Skill set: Protein purification LLPS assays Biochemistry: Exosome isolation Electron microscopy Light and Fluorescence microscopy Drosophila genetics Cell and tissue culture Neural stem cell culture Molecular Biology Fluorescence in situ hybridization Drug screening
