About Carl Diehl

I am currently employed as Senior Scientist at Saromics Biostructures. My main areas of expertise are NMR spectroscopy and protein expression and purification. In addition I also work with biophysical characterisation, fragment screening, crystallisation, system administration of the company network and programming to develop software for in-house applications. I have worked with NMR spectroscopy since my PhD and have experience within most areas of protein NMR. In my current position, I am mainly working on NMR characterisation of biosimilars using high-resolution NMR, fragment screening using ligand-detected and protein-detected techniques and structural characterisation of peptides and proteins including structure determinations in apo or complex with ligand. I have created numerous pipelines for handling the diverse sets of data within NMR. I have worked extensively with Weak Affinity Chromatography, a fragment screening technique that uses HPLC-MS, where I have played a major role in creating a pipeline where we can receive plates with unmixed fragments and deliver screening results within a week. Within the field of protein expression and purification, I have worked with everything from clone design, and molecular biology to protein expression and purification. I have mostly produced protein for crystallography, NMR spectroscopy and biophysical characterisation, where high purity of the protein is required. I am well versed in various chromatographic techniques for protein purification and in my current position I have purified protein from various sources including E.coli, insect and mammalian cells. My other areas of expertise are biophysical characterization and binding studies where I have worked with ITC, CD and fluorescence spectroscopy, thermal scanning fluorimetry (DSF), HPLC-MS, MST and DLS. Lately I have been going into implementing machine learning/AI techniques to interpret and predict results from NMR and fragment screens.
