Bridging the gap between computational and experimental biophysics to tackle the following:(1) The structural basis of the RNA chaperone activity of the bacterial Hfq protein. This project involves NMR spectroscopy, single molecule TIRF assays, developing a solution confocal single molecule FRET assay, native gel shift assays and all-atom MD simulations.(2) The basis of cooperatively, tertiary structure motifs and evolutionary constraints in the folding of structured RNAs (using the glmS ribozyme as a model). This project involves site-directed mutagenesis, gel-based activity assays and all-atom MD simulations.
