About Griffin Kramer
I'm a graduate of UCSB’s College of Creative Studies, with a research interest in genomics, transcriptomics, and computational biology. During my time at UCSB I conducted research in the Richardson Lab, where I worked on CRISPR interference (CRISPRi) off-target identification and mapping. I developed a computational pipeline, MOTREC, to analyze the data and provide a list of ranked on and off-target interactions between dCas9 and a eukaryotic genome. I used this pipeline to demonstrate that our off-target mapping technique also works on cutting guides, not just those used to knockdown genes. I compared our method to other off-target mapping methods that use ChIP-seq on MRE11, a gene involved in DNA double strand break repair. MOTREC output can be used to re-annotate initial CRISPRi screen data, making it a powerful tool for improving screen strength.
