Currently at UAB’s Flow Cytometry & Single Cell Services Core Facility, I am handling and managing very high end sate of arts flow cytometers such as BD Symphony A5 equipped with 6 different lasers, BD Symphony A3equipped with 5 different lasers, BD Fortessa, BD LSRII, and ThermoFisher’s Aquatics focusing flow cytometer Attune NxT, and sort samples on various BD ARIA sorters and on BD Melody sorters; acquire and analyze samples on Amnis ImageStream Imaging Flow Cytometer which allows simultaneously quantitative power of Flow Cytometry with High Content Image Analysis. In my previous research lab, I was studying the role of innate immune cells, particularly how neutrophils mediated chronic inflammation in different diseases such as HIV-1 infection, cancer, and most recent COVID-19. Using flow cytometric immunophenotyping we were characterizing unique neutrophils subpopulation present in low density neutrophils (LDNs) in the peripheral blood of these patients. Identification of disease linked neutrophil subpopulations and their mechanism of driving inflammation will provide diagnostic and therapeutic benefits. During PhD and as postdoctoral fellow I have studied the way cells transmit and meticulously execute the information to regulate cell growth, proliferation, and survival. Aberrations in executing these precise signals develop cancer. Understanding various molecular signatures to predict the mechanisms responsible for the exact cell related processes that may ultimately lead to the development of different strategies to prevent/cure the development of these inflammation related diseases. I am highly motivated to work on these challenges that could lead to the development of new strategies to combat these diseases. Scientific Skills and Experience: •Multi-color flow cytometry techniques, Multiplex cytokine assay, Microscopy, Live cell imaging and Western blotting. Working knowledge of the 10x Chromium, Fluidigm, and Rhapsody instruments related to single cell sequencing •Mammalian cell culture, isolation of primary and immune cells from tissues. •Innate immunity and inflammation, immune check point inhibitors and immune therapy. •CITEseq sample preparation for single cell mRNA and protein expression analysis. •Apoptosis, cell cycle analysis, cancer cell migration and invasion, EMT and metastasis. •Metabolomic-Sample preparation of metabolomics and Lipidomics, hand on analysis tool such as mzMIne, XCMS, MetaboAnalyst, Mummichog, Metabolome workbench and experience in data base searches.
