About Tashvinder Singh
I did my PhD thesis on "Metabolic Modulation of breast cancer cells using trehalose." My doctoral research focuses on alterations in mitochondrial metabolism in breast cancer and metabolic heterogeneity among low and high-metastatic breast cancer cells using a synergistic combination of dietary supplements Trehalose and targeted chemotherapy. I analyzed the differential metabolic demands of breast cancer subtypes and their heterogeneous response to therapeutic strategies involving a combination of therapies. My doctoral research work includes the use of therapeutic strategies involving trehalose as a metabolic modulatory dietary drug in combination with standard anti-cancer drugs. This includes analyzing basal OCR SUIT protocols for analyzing leak respiration mitochondrial membrane integrity coupling/uncoupling efficiency ETC complex activity etc. among different breast cancer cell types. Trehalose-mediated targeting of the PI3K/Akt pathway was found to be a promising supplementary therapy in combination with tamoxifen and colchicine against ER-positive breast cancer cells via suppressing NADPH-linked complex I activity and reducing the uncoupling efficiency of mitochondrial respiration. Beyond my primary research I have also been involved in drug discovery cytotoxicity studies and a project on bone marrow-derived stem cells analyzing their chondrogenic potential for treating knee defects. This work involved isolating mesenchymal stem cells chondrocytes and adipocytes and their 3D hydrogel-based cultures bioprinting and subsequent chondrogenic analysis (Articles published). I was also part of another study investigating the neuroprotective potential of Dapagliflozin an SGLT2 inhibitor against STZ-NAD-induced diabetic neuropathy (DN) in Wistar rats via IGF-1 signaling. The study involved the evaluation of DN through behavioral assessments following Dapagliflozin treatment and molecular analysis of the brain and sciatic nerve using western blotting qRT-PCR immunohistochemistry and histopathology (Published). Additionally I was part of a study focused on evaluating the association of risk factors and altered expression of RAAS genes in type 2 diabetes mellitus (T2DM) patients with and without complications (retinopathy nephropathy and neuropathy). The study involved the analysis of various risk factors and the expression of RAAS pathway genes (AGT ACE ACE2 and AGT1R) using qRT-PCR in healthy controls T2DM patients with complications and T2DM patients without complications (Published).
