Summary of TECHNICAL EXPERIENCE: In Vitro Assay Systems: • Immune cell isolation/activation/differentiation : Murine splenocytes, lymph node cells, fetal liver cells, and T cells, and Human PBMCs, monocytes, T cells, NK cells, and B cells. • Murine and human monocyte/macrophage, B and T cell, NK cell, PBMC, splenocytes, and whole blood stimulation: used numerous in vitro or ex vivo immune cell activation methods and measured proliferation, activation marker expression and cytokine/chemokine and antibody production. • Human and murine cytotoxicity assays: T cell, NK, ADCC • ELISAs: developed and employed ELISA based systems or utilized commercial kits to measure antibodies, cytokines or other soluble factors. • Protein/antibody radioligand binding assays to whole cell and cell membrane preparations: ligands included proteins, peptides and antibodies. • Flow cytometric analysis: fluorescent staining of cell surface and intracellular proteins, C1q deposition and apoptosis assays (FACSCalibur, Accuri). • Octet Bio-Layer Interferometry for the determination of antibody Kds • Chemotaxis assays: primary cells and cell lines (human, monkey, rabbit, dog and rodent). • Ca flux assay • Bactericidal culture techniques and assays: propagation, agar plate counts, complement deposition, phagocytosis, PI uptake. • Cell Culture: maintenance of a variety of cell lines and primary human and murine cell cultures, cell differentiation • Viral culture (HIV, EBV, CMV, vaccinia, etc.) • EBV transformation of B cells In Vivo techniques: • Murine and rat handling, injections (iv, sc, id, ip) and organ harvest (spleens, lymph nodes, etc.). • Murine Tumor immunotherapy studies: injection of various tumors (sc, ip), monitored tumor growth (caliper measurements), harvested tumors for immune-histochemistry, and harvested spleens and lymph nodes for various immune measurements. • Phlebotomy: chickens, goats, sheep, mice and rats
